In vivo testing typically starts with mice or rats, and then progresses up to human testing. A candidate drug must have favorable properties in each successive species before the next species is tested.

- In Vivo - Artemisinin - MODEL FOR THE ASSESSMENT OF CLINICALLY RELEVANT ANTIMALARIAL CROSS-RESISTAN CE - University of Vienna Austria AT - 2001: Cross-resistan ce decreases drug susceptibility of Plasmodium falciparum. This is to find out whether any clinically relevant cross-sensitiv ity of P. falciparum exists between artemisinin and mefloquine. 76 patients with falciparum malaria were treated with artemisinin derivatives. In vitro drug sensitivity tests were performed with artemisinin, mefloquine, quinine, and chloroquine. Distinct in vitro cross-sensitiv ity between artemisinin and mefloquine was observed1). To assess the relevance of this finding for clinical cross-resistan ce, we used an analytical model based on the relation of in vivo treatment response parameters (fever, parasite and symptom clearance) to a single reference drug with in vitro drug sensitivity data of several other drugs. Artemisinin and mefloquine in vitro drug sensitivities were equally well reflected in the in vivo treatment response to artemisinin, thereby suggesting the clinical relevance of in vitro cross-sensitiv ity.
- In Vivo - Artemisinin - Qinghaosu (artemisinin): an antimalarial drug from China - Science - CN - 1985: The herb Artemisia annua has been used for many centuries in Chinese traditional medicine as a treatment for fever and malaria. In 1971, Chinese chemists isolated from the leafy portions of the plant the substance responsible for its reputed medicinal action. This compound, called qinghaosu (QHS, artemisinin), is a sesquiterpene lactone that bears a peroxide grouping and, unlike most other antimalarials, lacks a nitrogen-conta ining heterocyclic ring system. The compound has been used successfully in several thousand malaria patients in China, including those with both chloroquine-se nsitive and chloroquine-re sistant strains of Plasmodium falciparum. Derivatives of QHS, such as dihydroqinghao su, artemether, and the water-soluble sodium artesunate, appear to be more potent than QHS itself. Sodium artesunate acts rapidly in restoring to consciousness comatose patients with cerebral malaria. Thus QHS and its derivatives offer promise as a totally new class of antimalarials.
